The cellular metabolic process whereby new strands of DNA are synthesized. The template for replication can either be an existing DNA molecule or RNA. The process of restoring DNA after damage. Genomes are subject to damage by chemical and physical agents in the environment (e.g. UV and ionizing radiations, chemical mutagens, fungal and bacterial toxins, etc.) and by free radicals or alkylating agents endogenously generated in metabolism. DNA is also damaged because of errors during its replication. A variety of different DNA repair pathways have been reported that include direct reversal, base excision repair, nucleotide excision repair, photoreactivation, bypass, double-strand break repair pathway, and mismatch repair pathway. Repair of a DSB made between two repeated sequences oriented in the same direction occurs primarily by the single strand annealing pathway. The ends of the break are processed by a 5' to 3' exonuclease, exposing complementary single-strand regions of the direct repeats that can anneal, resulting in a deletion of the unique DNA between the direct repeats. Synthesis of DNA that proceeds from the broken 3' single-strand DNA end uses the homologous intact duplex as the template. A DNA repair process in which that does not require the exchange of genetic material between the broken DNA molecule and a homologous region of DNA. The repair of double-strand breaks in DNA via homologous and nonhomologous mechanisms to reform a continuous DNA helix. The synthesis of DNA that contributes to the process of double-strand break repair via single-strand annealing.
