Catalysis of the reaction: cyclobutadipyrimidine (in DNA) = 2 pyrimidine residues (in DNA). The enzyme catalyzes the reactivation by light of irradiated DNA. A similar reactivation of irradiated RNA is probably due to a separate enzyme. Catalysis of the cleavage of C-C bonds by other means than by hydrolysis or oxidation, or conversely adding a group to a double bond. Catalysis of the cleavage of C-C, C-O, C-N and other bonds by other means than by hydrolysis or oxidation, or conversely adding a group to a double bond. They differ from other enzymes in that two substrates are involved in one reaction direction, but only one in the other direction. When acting on the single substrate, a molecule is eliminated and this generates either a new double bond or a new ring. Catalysis of the repair of a photoproduct resulting from ultraviolet irradiation of two adjacent pyrimidine residues in DNA. Catalysis of the reaction: pyrimidine-pyrimidone (6-4) photoproduct (in DNA) = 2 pyrimidine residues (in DNA). Catalyzes the reactivation of ultraviolet-irradiated DNA.
