Repair of a DSB made between two repeated sequences oriented in the same direction occurs primarily by the single strand annealing pathway. The ends of the break are processed by a 5' to 3' exonuclease, exposing complementary single-strand regions of the direct repeats that can anneal, resulting in a deletion of the unique DNA between the direct repeats. The removal of nonhomologous sequences at the broken 3' single-strand DNA end before DNA repair synthesis can occur. A process, occurring at the cellular level, that is involved in the reproductive function of a multicellular or single-celled organism. A DNA repair process in which that does not require the exchange of genetic material between the broken DNA molecule and a homologous region of DNA. The conversion of the mating-type locus from one allele to another resulting from the recombinational repair of a site-specific double-strand break at the mating-type locus with information from a silent donor sequence. There is no reciprocal exchange of information because the mating-type locus copies information from the donor sequence and the donor sequence remains unchanged. During DSBR via single-strand annealing, the removal of nonhomologous sequences at the broken 3' single-strand DNA end before DNA repair synthesis can occur. The chemical reactions and pathways resulting in the breakdown of DNA, involving the hydrolysis of internal 3',5'-phosphodiester bonds in one or two strands of deoxyribonucleotides. The repair of double-strand breaks in DNA via homologous and nonhomologous mechanisms to reform a continuous DNA helix.
